For several years I have been using Testsimplets, a prestained slide
from Boeringer-Mannheim, which stains the nuclei of leukocytes very
well, showing the definitive bridges between the clumps of chromatin.
Immature spermatocytes will not have those bridges. I also use the same
slide for sperm morphology determinations. Staining requires a 10 ul
drop of semen and takes 2-5 minutes at room temperature for the staining
to be complete. The one drawback, is that the staining is temporary,
lasting only overnight (if the slide is refrigerated). I'd be very
interested to hear what others are doing in this area of investigation.
