Routine staining to differentiate round cells in semen

Dear Colleagues,

I would like to employ staining to differentiate round cells in semen in my
clinical Andrology lab, but I have been hesitant to do so because of the
toxicity of the reagents. The techniques I am aware of are the benzidine-
cyanosine and the o-toluidine blue staining methods. Both of these involve
known carcinogens. Do some Andrology labs use these routinely, and, if so,
what safety precautions are used? Alternatively, do any of you know of a
less toxic method that can be performed in the time-frame required for
clinical Andrology (e.g., we often wish to detect leukocytes before choosing
to do a simple wash to prepare oligozoospermic specimens for IUI).
